Properties and Biology
BBa_K4467002 is a protein coding sequence comprising the small fungal effector protein, Pst_12806, present in the haustorium-of the wheat stripe rust fungus, Puccinia striiformis f.sp.tritici (Pst). Upon infection, it is translocated into chloroplast and affects chloroplast function (Xu,Q., 2019). It suppresses plant basal immunity by reducing callose deposition and the expression of defense-related genes (Xu,Q., 2019). Expression of Pst_12806 in plants reduces electron transport rate, photosynthesis, and production of chloroplast-derived ROS (Xu,Q., 2019).
Its knockdown (by host-induced gene silencing) reduces Pst growth and development, likely due to increased ROS accumulation. Pst_12806 interacts with the C-terminal Rieske domain of the wheat TaISP protein.
Usage
As part of the 2022 UNSW_Australia project to design a peptide based inhibitor for Puccinia sp., we used Pst_12806 as a potential target. This protein was expressed in NEB TURBO and NEB T7 express E.coli cells. Its binding with TaiSP partner protein was analysed using FRET based interaction analysis.
